Assassin Missions

Do these count towards the story mode? Im at 55% complete, 83 missions done but I now have no1 left on the map to get jobs from bar the assassin mission.

sorry for the off topic but are the assassin missions in anyway simular to Saints Row where you must manually hunt down your target? or are they immediatly given to you on the radar?
