Yes, if you're perfect. Otherwise you'll have to replay some levels, but it's not a big deal.

What if I just play the story, not caring about symbols or questions - can I go back and do "clean up" and get these last things later (like flags in AssCreed for example)?

I'd rather just play for the story and do that stuff afterwards, if possible.

I don't think this would be a problem, but unlike Assassins Creed you're not going to want to play this anymore than you have to!
