I have seen a lot of confusion on these forums as to what are these "glyphs" people are referring to and how to get the achievements related to them. I shall therefore try to clear up this confusion with this mini-guide.
"Glyphs" are basically imprints left by Subject 16. Subject 16, like Desmond, was imprisoned by Abstergo. He left behind these imprints so the next person in the Animus could make sense of all of it. These glyphs have the videos locked up, and it is your job to find them all and decode them. This also ties in three achievements, Tip of the Iceberg, A Piece of the Puzzle, and Vitruvian Man.
The way the glyph works is once you find it, you turn eagle vision on and go "read" it by pressing the "y" button. This unlocks the Tips of the Iceberg achievement. After that, you have to decode a small puzzle. Once you decode it, you earn A Piece of the Puzzle achievement and obtain a small segment of a video. If you collect all 20 segments of the video you can watch it all and better understand your role in all of Assassins Creed 2. You also earn Vitruvian Man once you collect all of the pieces of the video.
So now that you know a little bit about these glyphs you may be wondering how to find them. Well, you are first introduced to them by Shawn when you arrive at the Villa. If you turn Eagle Vision on and go to the left side of the front of the Villa you will see a white spot. Climb to it and you find your first glyph. You then have to decode a puzzle. I am not sure if the puzzle's are set or if they are random, however they are not very difficult so I will not write how to solve them all as I haven't found all of them yet.
Anyways, the rest of the glyphs are scattered all around the landmarks of major cities. If you climb all the viewpoints in a particular city you will see all the major landmarks in the said city. If you go to the Pause menu, and then go to Database > Locations, you will then see cities and the landmarks in them in a list. If you go to the list, and scroll through the pictures, you will notice some of them have a red symbol on them. This means that a glyph is somewhere close to that particular landmark. When you look for the glyph, you must locate "the eye". It will look something similar to this.

Once you find the eye, turn eagle vision on and repeat the process. The eye is sometimes difficult to find, but it is usually either going to be inside the building if it is some sort of courtyard or on the roof of the building.
